Studie Doelstellingen:
- Specifieke veranderingen identificeren in de expressie van boodschapperribonucleïnezuur (mRNA)/microribonucleïnezuur (miRNA) in spieren geassocieerd met hogere of lagere relatieve maten van mitochondriale capaciteit en vetoxidatie.
- Om uitgescheiden factoren/miRNA's te identificeren die specifiek betrekking hebben op de metabole respons van spieren en die aanwezig zijn na een enkele eerste inspanning.
- De juiste klinische monsters verzamelen (spier- en vetweefsel, plasma/serum) om validatie mogelijk te maken van myokines geassocieerd met veranderingen in zuurstofverbruik/mitochondriale inhoud via in vivo en in vitro ontdekkingspogingen.

Geldig voor alle Groepen
- Gezonde mannen en vrouwen van 18 tot en met 40 jaar.
- Bereid om alcohol- en cafeïneconsumptie te stoppen gedurende 48 uur voorafgaand aan elke bloedafname
Geldt voor Groep 1
- BMI tussen 22 en 29,9 kg/m2
- Niet betrokken bij regelmatig oefenprogramma
- Bereid om gedurende de studieperiode elke dag te sporten
Geldt voor Groep 2
- BMI tussen 22 en 29,9 kg/m2
Maximale zuurstofopname (VO2max) ≥ 45 ml/kg vetvrije massa

- Minimaal 1,5 uur aerobe training met matige tot zware intensiteit, 3 keer per week

Uitsluitingscriteria:
Van toepassing op alle groepen
- Geschiedenis van diabetes type 2
- "Ongunstige anatomie" voor continue veneuze bloedafname
- Abnormaal rust-ECG
- Significante nier-, hart-, lever-, long- of neurologische ziekte (gecontroleerde hypertensie is acceptabel als baseline bp < 140/90 op medicijnen)
- Gebruik van geneesmiddelen waarvan bekend is dat ze het energiemetabolisme of lichaamsgewicht beïnvloeden: inclusief, maar niet beperkt tot: orlistat, sibutramine, efedrine, fenylpropanolamine, corticosteron, enz.
- Huidige behandeling met bloedverdunners of bloedplaatjesaggregatieremmers die niet veilig kunnen worden gestopt voor testprocedures
- Nieuw begin (<3 maanden op een stabiel regime) gebruik van orale anticonceptiva of hormoonvervangingstherapie
- Alcohol- of ander drugsmisbruik
- Roken in de afgelopen 3 maanden
- Vrouwtjes die momenteel zwanger zijn of zijn geweest of die momenteel een kind hebben of hebben verzorgd in de afgelopen 12 maanden (minimum).
- Ouderlijke deelname aan de studie die het welzijn van het kind in gevaar brengt [geen partner of verbonden verzorger]
- Niet bereid of niet in staat om zich te onthouden van cafeïne of alcohol 48 uur voorafgaand aan metingen van de stofwisseling
- Verhoogde leverfunctietesten
- Metalen voorwerpen die de meting van de lichaamssamenstelling/magnetische resonantiespectroscopie zouden kunnen verstoren, zoals geïmplanteerde staafjes, chirurgische clips, enz.
- Elke New York Heart Association-klasse van congestief hartfalen
- Geschiedenis van diepe veneuze trombose of longembolie
- Aanzienlijke spataderen
- Abnormaal aantal bloedcellen/bloedarmoede of bloeddonatie in de afgelopen 2 maanden
- Grote operatie aan de buik, het bekken of de onderste ledematen in de afgelopen 3 maanden
- Bariatrische chirurgie of liposuctie in de afgelopen 3 jaar
- Kanker (actieve maligniteit met of zonder gelijktijdige chemotherapie)
- Reumatoïde ziekte
- Bypass-transplantaat in ledemaat
- Bekende genetische factor (Factor V Leiden, enz.) of hypercoaguleerbare toestand
- Gediagnosticeerde perifere arteriële of vasculaire ziekte, of claudicatio intermittens
- Familiegeschiedenis van primaire diepe veneuze trombose of longembolie
- Perifere neuropathie
- Claustrofobie
- Frequent nachtelijk urineren en/of slaapapneu
- Aanwezigheid van een aandoening die, naar de mening van de onderzoeker, de veiligheid van de deelnemer of de gegevensintegriteit of het vermogen van de deelnemers om het trainingsprotocol te voltooien in gevaar brengt

Experimenteel: Groep 1 - Regelmatige lichaamsbeweging
Wissel intervaltraining en aerobe training en lichaamsbeweging af

Een warming-up van 5 minuten en een cooling-down van 5 minuten respectievelijk voor en na elke trainingssessie.
Er zullen afwisselende dagen intervaltraining en aerobictraining zijn.
De intervaltraining wordt uitgevoerd op een hometrainer, terwijl de aerobe training wordt uitgevoerd op een loopband.
De intervaltraining bestaat uit periodes van 5 minuten met een hogere intensiteit afgewisseld met 4 minuten met een lagere intensiteit voor een totale duur van 45 minuten.
De intensiteit neemt elke week toe.
De aerobe trainingscomponent zal worden vastgesteld op een matige intensiteit, maar zal in de derde en laatste week wekelijks toenemen van 45 minuten naar 75 minuten naar 90 minuten.

Het verschil wordt gemeten bij zwaarlijvige, magere en atletische deelnemers. De hersteltijdconstante van Phosphocreatine (PCr) en het PCr-niveau in zuurstofrijke spieren in rust zullen worden gebruikt om de maximale mitochondriale capaciteit te berekenen. |
