MÅL: I. Sammenlign lokoregional kontrol, sygdomsfri overlevelse og overordnet overlevelse hos patienter med carcinom i prostata, som anses for at have høj risiko for tilbagefald og modtager langtidsadjuverende zoladex (ZDX) vs. ingen adjuverende behandling efter neoadjuverende ZDX/ flutamid og strålebehandling. II. Sammenlign seksuel funktion hos patienter behandlet med disse to regimer.
OVERSIGT: Randomiseret undersøgelse. Arm I: Neoadjuverende antiandrogenterapi og frigørende faktor agonistterapi plus strålebehandling. Flutamid, FLUT, NSC-147834; og Zoladex, ZDX, NSC-606864; plus ekstern strålebestråling ved hjælp af megaspændingsudstyr med energier på mindst 4 MV (mindst 6 MV foretrækkes). Arm II: Neoadjuverende antiandrogenterapi og frigørende faktor agonistterapi plus strålebehandling efterfulgt af adjuverende frigørende faktor agonistterapi. FLUT; og ZDX; plus ekstern strålebestråling ved hjælp af udstyr som i arm I; efterfulgt af ZDX.
PROJEKTERET TILSLUTNING: 1.508 patienter vil blive optjent over 2 år.

SYGDOMSKARAKTERISTIKA: Histologisk bekræftet, lokalt fremskreden adenocarcinom i prostata, inklusive: Voluminøse primære tumorer begrænset til prostata (klinisk stadie T2c) Primære tumorer, der strækker sig ud over kapslen (klinisk stadie T3-4) Ingen almindelig involvering af iliaca eller para-aortisk nodal lymfeknudeinvolvering under det almindelige iliaca-niveau tilladt.
