ZIELE: I. Vergleich der lokoregionären Kontrolle, des krankheitsfreien Überlebens und des Gesamtüberlebens bei Patienten mit Prostatakarzinom, bei denen ein hohes Rückfallrisiko gilt und die langfristig eine adjuvante Behandlung mit Zoladex (ZDX) erhalten, im Vergleich zu keiner adjuvanten Behandlung nach neoadjuvanter ZDX/ Flutamid und Strahlentherapie. II. Vergleichen Sie die sexuelle Funktion bei Patienten, die mit diesen beiden Therapien behandelt wurden.
ÜBERBLICK: Randomisierte Studie. Arm I: Neoadjuvante Antiandrogentherapie und Releasing-Faktor-Agonisten-Therapie plus Strahlentherapie. Flutamid, FLUT, NSC-147834; und Zoladex, ZDX, NSC-606864; plus externe Bestrahlung mit Megaspannungsgeräten mit Energien von mindestens 4 MV (mindestens 6 MV bevorzugt). Arm II: Neoadjuvante Antiandrogentherapie und Releasing-Faktor-Agonisten-Therapie plus Strahlentherapie, gefolgt von einer adjuvanten Releasing-Faktor-Agonisten-Therapie. FLUT; und ZDX; plus externe Bestrahlung mit Geräten wie in Arm I; gefolgt von ZDX.
PROJEKTIERTE ACCCRUALITÄT: 1.508 Patienten werden über einen Zeitraum von 2 Jahren angesammelt.

EIGENSCHAFTEN DER KRANKHEIT: Histologisch bestätigtes, lokal fortgeschrittenes Adenokarzinom der Prostata, einschließlich: Massenhafter Primärtumoren, die auf die Prostata beschränkt sind (klinisches Stadium T2c) Primärtumoren, die sich über die Kapsel hinaus erstrecken (klinisches Stadium T3-4) Keine häufige Beteiligung des Becken- oder paraaortalen Knotens. Regional Lymphknotenbeteiligung unterhalb der Ebene des gemeinsamen Beckens zulässig. Positive Knoten in bildgebenden Untersuchungen müssen durch FNA oder chirurgische Probenahme biopsiert werden. PSA nicht mehr als 150 (obligatorisch). Keine Fernmetastasen
