Un essai clinique interventionnel adaptatif de phase 3b pour évaluer l'efficacité et l'innocuité du tralokinumab 300 mg toutes les deux semaines en monothérapie par rapport à un placebo chez des sujets atteints d'eczéma atopique modéré à sévère des mains qui sont candidats à un traitement systémique

Le but de cette étude est de tester si le traitement par tralokinumab est sûr et efficace pour traiter l'eczéma atopique modéré à sévère des mains. Cela sera jugé par une série d'évaluations qui évaluent la gravité et l'étendue de l'eczéma atopique des mains et de ses symptômes, ainsi que l'état de santé général et la qualité de vie.

L'essai durera jusqu'à 40 semaines. Il y aura jusqu'à 15 visites, dont 3 seront effectuées par téléphone. La première partie de l'essai s'appelle une période de dépistage et durera jusqu'à 4 semaines. Pendant les 16 premières semaines après le dépistage, les participants à l'essai recevront du tralokinumab ou des injections factices toutes les deux semaines. Critère d'intégration:

  • Âge de 18 ans ou plus au moment du dépistage
  • Diagnostic de la dermatite atopique (DA) selon les critères de Hanifin et Rajka (1980) pour la DA
  • Antécédents de MA depuis ≥ 1 an
  • Présence d'eczéma atopique des mains (AHE) qui a persisté pendant plus de 3 mois ou qui est revenu deux fois ou plus au cours des 12 derniers mois, en évitant tout irritant et allergène connu et pertinent
  • Atteinte de la MA d'au moins une partie du corps autre que les mains et les poignets au moment du dépistage
  • Score AHE d'évaluation globale de l'investigateur de 3 ou 4 (modéré à sévère) au dépistage et au départ
  • Un score de démangeaison HESD (moyenne hebdomadaire) ≥ 4 au départ
  • Sujets qui ont des antécédents récents documentés (dans les 12 mois précédant la visite de dépistage) de réponse inadéquate au traitement de l'AHE avec des médicaments topiques sur ordonnance ou pour lesquels les traitements topiques sont autrement médicalement déconseillés (par exemple, en raison d'effets secondaires importants ou de risques pour la sécurité)
