Eine offene Phase-Ia/Ib-Dosiseskalationsstudie zur Bewertung der Sicherheit und Pharmakokinetik von ROSE12 als Einzelwirkstoff und in Kombination mit anderen Antitumorwirkstoffen bei Patienten mit lokal fortgeschrittenen oder metastasierten soliden Tumoren
Hierbei handelt es sich um eine offene Dosissteigerungsstudie der Phase Ia/Ib zur Bewertung der Sicherheit und Pharmakokinetik von ROSE12 als Einzelwirkstoff und in Kombination mit anderen Antitumorwirkstoffen bei Patienten mit lokal fortgeschrittenen oder metastasierten soliden Tumoren.
Die Studie wird aus drei Teilen bestehen: einem Dosissteigerungsteil, einem Biopsieteil (der Teil zur Bewertung von Biomarkern) und einem Erweiterungsteil.

Einschlusskriterien:
- Alter ≥ 18 Jahre zum Zeitpunkt der Unterzeichnung der Einwilligungserklärung (ICF)
- Eastern Cooperative Oncology Group (ECOG) PS von 0 oder 1
- Ausreichende hämatologische und Endorganfunktion
- Lebenserwartung ≥ 12 Wochen
- Patienten mit histologischer Dokumentation eines lokal fortgeschrittenen oder metastasierten soliden Tumors
- [Dosiseskalationsteile und Expansionsteil] Patienten mit bestätigter Verfügbarkeit von frischem Tumor oder repräsentativen Tumorproben
- [Biopsieteile] Patienten mit zugänglichen Läsion(en)
Ausschlusskriterien:
- Klinisch signifikante Herz-Kreislauf- oder Lebererkrankung
- Behandlung mit einer Prüftherapie und einer Krebstherapie innerhalb von 28 Tagen vor Beginn der Studienmedikation
- Jegliche Vorgeschichte eines immunvermittelten unerwünschten Ereignisses vom Grad 4, das auf eine frühere Krebsimmuntherapie zurückzuführen ist (mit Ausnahme einer asymptomatischen Erhöhung der Serumamylase oder Lipase).
- Alle imAEs aus einer früheren Krebsimmuntherapie (außer Endokrinopathie, die mit einer Ersatztherapie behandelt wird, stabiler Vitiligo oder stabiler Alopezie), die nicht vollständig auf den Ausgangswert zurückgegangen sind.
- Unerwünschte Ereignisse aus einer früheren Krebstherapie, die nicht auf Grad ≤ 1 abgeklungen sind, mit Ausnahme von Alopezie, Vitiligo oder Endokrinopathie, die mit einer Ersatztherapie behandelt werden
- Primäre Malignität des Zentralnervensystems (ZNS), unbehandelte ZNS-Metastasen, die eine Antitumorbehandlung erfordern, oder aktive ZNS-Metastasen
- Unkontrollierter tumorbedingter Schmerz
- Unkontrollierter Pleuraerguss, Perikarderguss oder Aszites, der wiederholte Drainageverfahren erfordert
- Aktive oder Vorgeschichte einer klinisch signifikanten Autoimmunerkrankung
- Schwere allergische, anaphylaktische oder andere Überempfindlichkeitsreaktionen auf chimäre oder humanisierte Antikörper oder Fusionsproteine in der Vorgeschichte.
