Deze klinische studie zal de veiligheid en immunogeniciteit beoordelen van 3 dosisniveaus van het mRNA-1647 cytomegalovirusvaccin bij CMV-seronegatieve en CMV-seropositieve gezonde volwassenen van 18-40 jaar oud.

mRNA-1647-P202 is een 2-delige studie.
Deel 1 van de studie evalueert de veiligheid en immunogeniciteit van lage, gemiddelde en hoge dosisniveaus van mRNA-1647-vaccin of placebo, toegediend volgens een schema van 0, 2, 6 maanden bij gezonde CMV-seronegatieve en CMV-seropositieve mannen en vrouwen , 18 tot 40 jaar.
Een geplande tussentijdse analyse van veiligheid en immunogeniciteit tot en met maand 3 (1 maand na de tweede dosis) van deel 1 van het onderzoek vormde de basis voor de selectie van het middelste dosisniveau voor verdere ontwikkeling.
Deel 2 van de studie is opgezet om de veiligheid en immunogeniciteit van het middelste dosisniveau van het mRNA-1647-vaccin of placebo verder te evalueren volgens een schema van 0, 2, 6 maanden bij ongeveer 200 gezonde deelnemers van 18 tot 40 jaar oud, bestaande uit: CMV-seronegatieve en CMV-seropositieve vrouwelijke populatie, waaronder de doelpopulatie voor de cruciale fase 3-werkzaamheidsstudie.

Inclusiecriteria:
- Man of vrouw 18-40 jaar (Deel 1); Vrouw 18-40 jaar (Deel 2)
- Begrijpt en stemt ermee in zich te houden aan de procesprocedures en geeft schriftelijke geïnformeerde toestemming
- Naar het oordeel van de Onderzoeker verkeert hij in een goede algemene gezondheid en is hij in staat om de procesprocedures na te leven
- Body mass index (BMI) 18-35 kilogram/meter (kg/m^2)
- Vrouwelijke deelnemers moeten ofwel niet vruchtbaar zijn ofwel aanvaardbare anticonceptiemethoden gebruiken vanaf ten minste 28 dagen voorafgaand aan de eerste vaccinatie en tot 3 maanden na de laatste vaccinatie en geven geen borstvoeding.
- Mannelijke deelnemers moeten ermee instemmen om adequate anticonceptie toe te passen vanaf het moment van de eerste vaccinatie en tot 3 maanden na de laatste vaccinatie.
Uitsluitingscriteria:
- Acuut ziek of koortsig op de dag van de eerste vaccinatie
- Voorafgaande ontvangst van een CMV-vaccin
- Abnormale resultaten van veiligheidslaboratoriumtesten
- Diagnose of aandoening die, naar het oordeel van de onderzoeker, klinisch onstabiel is of van invloed kan zijn op de veiligheid van de deelnemer, de beoordeling van veiligheidseindpunten, de beoordeling van de immuunrespons of het naleven van proefprocedures
- Heeft ≤28 dagen voorafgaand aan de eerste vaccinatie een vaccin gekregen of is van plan dit te krijgen of is van plan om binnen 28 dagen voorafgaand aan of na een studievaccinatie een niet-onderzoeksvaccin te krijgen, met uitzondering van een goedgekeurd griepvaccin dat >14 dagen ervoor kan worden toegediend of na een studievaccinatie. COVID-19-vaccins (ongeacht de fabrikant) kunnen >7 dagen maar bij voorkeur >14 dagen vóór of na een onderzoeksvaccinatie worden toegediend, met de bedoeling om COVID-19-vaccinatie voorrang te geven boven alle andere overwegingen.
- Voorafgaande ontvangst van chronische systemische immunosuppressiva of immuunmodificerende geneesmiddelen
- Ontvangst van intraveneuze immunoglobulinen of plasmaproducten binnen 3 maanden voorafgaand aan de dag van de eerste studievaccinatie
- Eerdere ontvangst van medicijnen in formulering van lipide nanodeeltjes (LNP) (alleen deelnemers van deel 1)
- Heeft ≥450 milliliter (ml) bloedproducten gedoneerd binnen 28 dagen na het screeningsbezoek
- Deelgenomen aan een interventioneel klinisch onderzoek binnen 28 dagen voorafgaand aan de dag van inschrijving
- Is een direct familielid of lid van het huishouden van het onderzoekspersoneel

Solicited ARs were selected signs and symptoms occurring after vaccination administration during a specified post-vaccination follow-up period.
The occurrence and intensity of the selected signs and symptoms was actively solicited from the participant during a specified post-vaccination follow-up period (day of vaccination and 6 subsequent days), using a predefined checklist in the electronic diary.
The following local ARs were solicited: pain at injection site, erythema (redness) at injection site, swelling/induration (hardness) at injection site, and localized axillary swelling or tenderness ipsilateral to the vaccination arm.
The following systemic ARs were solicited: headache, fatigue, myalgia (muscle aches all over the body), arthralgia (aching in several joints), nausea/vomiting, rash, fever, and chills.
