Kuvaus
Sisällyttämiskriteerit:
- Osallistujat, jotka ovat terveitä lääketieteellisen arvioinnin mukaan, mukaan lukien sairaushistoria, fyysinen tutkimus ja laboratoriotutkimukset.
- Painoindeksi (BMI) 18-30 kilogrammaa neliömetriä kohden (kg/m^2) (mukaan lukien) enintään 220 paunaan.
Sekä miehet että naiset ovat oikeutettuja osallistumaan seuraavilla tavoilla:
a. Naispuoliset osallistujat, jotka voivat fyysisesti tulla raskaaksi, joilla on vähintään yksi negatiivinen raskaustesti seulonnan aikana, ilmoittautumispäivänä, ennen tutkimustuotteen (IP) antoa, ennen CHM:ää ja malariahoidon alussa ja jotka suostuvat käyttämään tehokasta ehkäisyä raskauden välttämiseksi 28 päivää ennen ilmoittautumista koekäyntien loppuun asti.
- Pystyy antamaan allekirjoitetun tietoisen suostumuksen, joka sisältää tietoisessa suostumuslomakkeessa (ICF) ja tässä pöytäkirjassa lueteltujen vaatimusten ja rajoitusten noudattamisen sekä ymmärtämistestin suorittamisen, jos hän voi osallistua CHMI-menettelyyn.

Poissulkemiskriteerit:
- Akuutti sairaus tai kuume ≥99,5 Fahrenheit (F) (tai > 37,5 celsiusastetta) annostuspäivänä.
- Naiset, jotka ovat raskaana tai imettävät.
- Todisteet ja/tai historia kliinisesti merkittävistä lääketieteellisistä tiloista, kuten tutkijan arvioi, mukaan lukien pahanlaatuiset kasvaimet, diabetes mellitus ja epästabiili tai hallitsematon verenpaine.
- Viiden vuoden kardiovaskulaarinen riski >10 % Gazianon nomogrammia käytettäessä.

2 vartijaosapuolta satunnaistetaan suhteessa 1:1 saamaan MAM01 1,5 milligrammaa kilogrammaa kohti (mg/kg) IV tai lumelääkettä.
Vähintään 24 tunnin turvallisuusarviointijakson jälkeen 6 jäljellä olevaa kohortin 1 osallistujaa satunnaistetaan suhteessa 5:1 saamaan MAM01 1,5 mg/kg IV tai lumelääkettä.

Solicited AEs were defined events participants were specifically asked about, which were recorded by participants in the memory aid card.
Solicited AES included local injection site AEs (pain, redness, swelling, itching and bruising) and systemic AEs (fever, chills, headache, fatigue, nausea, muscle pain and joint pain).
A Solicited AE does not necessarily have a causal relationship with the intervention.

The characterization of protective efficacy against Pf following CHMI challenge, was assessed by evaluating the presence or absence of Pf infection as determined by qRT-PCR after CHMI (planned through 4 weeks post-CHMI) and evaluating the time to parasitemia after CHMI in each cohort.

Parasitemia was assessed by qRT-PCR up to Day 27 following CHMI.
In addition, a thick blood smear was prepared for microscopic analysis, which was examined only once the first qRT-PCR sample tested positive.
Daily parasitemia monitoring continued until the participant had a confirmed initial positive qRT-PCR.
The first positive qRT-PCR triggered either a second PCR test or microscopic analysis of the blood smear.
Once two positive results were obtained, rescue therapy was initiated.
