Einschlusskriterien:

  • Teilnehmer, die laut ärztlicher Untersuchung einschließlich Anamnese, körperlicher Untersuchung und Labortests gesund sind.
  • Body-Mass-Index (BMI) 18 bis 30 Kilogramm pro Quadratmeter (kg/m^2) (einschließlich) bis maximal 220 Pfund.
  • Teilnahmeberechtigt sind sowohl Männer als auch Frauen nach Maßgabe der folgenden Voraussetzungen:

    A. Weibliche Teilnehmerinnen, die körperlich schwangerschaftsfähig sind, haben während des Screenings, am Tag der Einschreibung, vor der Verabreichung des Prüfpräparats (IP), vor der CHM und zu Beginn der Malariabehandlung mindestens einen negativen Schwangerschaftstest und sind damit einverstanden, eine wirksame Empfängnisverhütung anzuwenden um eine Schwangerschaft ab 28 Tagen vor der Einschreibung bis zum Abschluss der Probebesuche zu vermeiden.

  • Kann eine unterzeichnete Einverständniserklärung abgeben, die die Einhaltung der in der Einverständniserklärung (ICF) und in diesem Protokoll aufgeführten Anforderungen und Einschränkungen sowie den Abschluss eines Verständnistests umfasst, wenn er/sie am CHMI-Verfahren teilnehmen darf.
  • Der gemeldete Abschluss der primären Coronavirus-Impfstoffserie (COVID) ist dokumentiert.

Ausschlusskriterien:

  • Akute Erkrankung oder Fieber ≥99,5°Fahrenheit (F) (oder >37,5 Grad Celsius) am Tag der Dosierung.
  • Frauen, die schwanger sind oder stillen.
  • Hinweise und/oder Anamnese klinisch bedeutsamer Erkrankungen nach Einschätzung des Prüfarztes, einschließlich bösartiger Erkrankungen, Diabetes mellitus und instabiler oder unkontrollierter Hypertonie.
  • Ein 5-Jahres-Kardiovaskuläres Risiko von >10 % anhand des Gaziano-Nomogramms.
  • Vorgeschichte einer Autoimmunerkrankung oder Immunschwäche oder einer anderen Beeinträchtigung des Immunsystems, einschließlich, aber nicht beschränkt auf das humane Immundefizienzvirus (HIV), Autoimmunerkrankungen oder immunsuppressive Therapie.
  • Teilnahme an einer interventionellen klinischen Studie und/oder Erhalt eines Prüfpräparats innerhalb von 180 Tagen vor der Verabreichung des Prüfpräparats am ersten Tag.
  • Voraussichtliche Einnahme von Arzneimitteln, von denen bekannt ist, dass sie Arzneimittelreaktionen mit Chloroquin oder Atovaquonproguanil (Malarone) hervorrufen, wie z. B. Cimetidin, Metoclopramid, Antazida und Kaolin.

2 Sentinel-Teilnehmer werden im Verhältnis 1:1 randomisiert und erhalten MAM01 1,5 Milligramm pro Kilogramm (mg/kg) intravenös oder Placebo. Nach einem mindestens 24-stündigen Sicherheitsüberprüfungszeitraum werden die 6 verbleibenden Teilnehmer der Kohorte 1 im Verhältnis 5:1 randomisiert und erhalten MAM01 1,5 mg/kg i.v. oder Placebo.

Parasitemia was assessed by qRT-PCR up to Day 27 following CHMI. In addition, a thick blood smear was prepared for microscopic analysis, which was examined only once the first qRT-PCR sample tested positive. Daily parasitemia monitoring continued until the participant had a confirmed initial positive qRT-PCR. The first positive qRT-PCR triggered either a second PCR test or microscopic analysis of the blood smear. Once two positive results were obtained, rescue therapy was initiated.
