本研究是一项多中心、开放标签的 I 期研究,剂量递增:2 个剂量水平。

纳入标准:
- 男性或女性患者 > 18 岁
组织学或细胞学(在复发的情况下)证实的结缔组织肿瘤,包括以下任何亚型:
- 脂肪肉瘤
- 纤维肉瘤、粘液纤维肉瘤
- 未分化多形性肉瘤
- 平滑肌肉瘤
- 仅多形性横纹肌肉瘤
- 血管肉瘤
- 分化不明的肿瘤:滑膜肉瘤、上皮样肉瘤、肺泡肉瘤、透明细胞肉瘤。
或骨肉瘤诊断,软骨肉瘤或脊索瘤。
- 未接受过放疗的局部晚期或局部复发的不能手术的肿瘤[不能手术的状态必须由工作人员评估,包括专门从事肉瘤的外科医生]。
- 先前未接受苹果酸舒尼替尼治疗
- 预期寿命 > 6 个月
- ECOG 体能状态 ≤ 2
血液检查、肾功能和肝功能在正常范围内,在进入研究前 7 天,血液或血清值如下:

排除标准:
- GIST、尤文肉瘤或胚胎性横纹肌肉瘤
- 辐射场包括肺、肠或中枢神经系统
- 预先存在的甲状腺异常,定义为尽管服药但甲状腺功能测试异常
- 研究药物给药前过去 4 周内出现 NCI ≥ 3 级出血
- 重大心血管疾病(纽约心脏协会(NYHA)>2级充血性心力衰竭、入组前6个月内发生心肌梗死、不稳定型心绞痛、严重心律失常、入组前6个月内发生严重脑血管意外、严重血栓栓塞病史(肺栓塞或深静脉血栓形成 DVT)入组前 6 个月内(最近有 DVT 病史且至少 6 周接受抗凝剂治疗(治疗性华法林除外)的患者符合资格)、QTc 间期延长(Bazett 时 QTc > 480 毫秒)、心动过缓(心率 < 45bpm)、电解问题(低钠血症 <120 毫摩尔/升、血钾≥6 毫摩尔/升)或在接受适当药物治疗时未控制的高血压(≥ 160 毫米汞柱收缩压和/或 ≥ 90 毫米汞柱舒张压)。
