Eine randomisierte, beobachterblinde Phase-2-Dosis-Wirkungs-Studie zur Bewertung der Sicherheit und Immunogenität von zwei verschiedenen Dosierungen eines abgeschwächten Lebendimpfstoffkandidaten gegen das Chikungunya-Virus (VLA1553) bei gesunden Kindern im Alter von 1 bis 11 Jahren

Hierbei handelt es sich um eine multizentrische, prospektive, randomisierte, beobachterverblindete, dreiarmige klinische Phase-2-Studie zur Bewertung der Volldosisformulierung von VLA1553, der Halbdosisformulierung von VLA1553 und der Kontrolle.

Detaillierte Beschreibung

Hierbei handelt es sich um eine multizentrische, prospektive, randomisierte, beobachterverblindete, dreiarmige klinische Phase-2-Studie zur Bewertung der Volldosis-Formulierung von VLA1553, der Halbdosis-Formulierung von VLA1553 und der Kontrolle (Nimenrix, ein tetravalenter Meningokokken-Impfstoff – Men ACWY).

Es werden mindestens 300 männliche und weibliche gesunde Kinder im Alter von 1 bis 11 Jahren eingeschrieben und die Gesamtverteilung der Teilnehmer beträgt 2:2:1 auf die beiden VLA1553-Dosisgruppen (jeweils n=120) oder die Kontrollgruppe (Nimenrix) (n=60). ).

Aus Sicherheitsgründen werden die ersten 30 Sentinel-Teilnehmer offen und nach einem Altersabstufungsschema in die Studie aufgenommen.

Nach der Sentinel-Analyse werden die Teilnehmer blind und randomisiert in drei Studienarme aufgenommen. Innerhalb jedes Behandlungsarms werden die Teilnehmer in drei Altersschichten eingeteilt:

Schicht A: 7 bis 11 Jahre – Kinder vom 7. Geburtstag bis zum Tag vor ihrem 12. Geburtstag.

Einschlusskriterien:

  1. Männliche oder weibliche gesunde Kinder im Alter von 7 bis 11 Jahren12 für Stratum A, 3 bis 6 Jahren13 für Stratum B und 1 bis 2 Jahren14 für Stratum C zum Zeitpunkt der Impfung;
  2. Schriftliche Einverständniserklärung der Eltern/rechtlich zulässigen Vertreter des Teilnehmers ((LAR(s)) entsprechend den örtlichen Anforderungen und ggf. schriftliche Einverständniserklärung des Teilnehmers;

Ausschlusskriterien:

  1. Teilnehmer mit IgM+/IgG- sind nicht für die Teilnahme an dieser Studie qualifiziert.
  2. Der Teilnehmer nimmt Medikamente oder eine andere Behandlung gegen ungelöste Symptome ein, die auf eine frühere CHIKV-Infektion zurückzuführen sind. oder an einer klinischen Studie mit einem CHIKV-Prüfimpfstoff teilgenommen hat;
