M-PRP Cellular Content Filgrastim과 Pegfilgrastim의 질과 양 비교 (M-PRP)
Filgrastim과 Pegfilgrastim을 사용하여 전구 세포의 말초 동원 후 수확된 M-PRP의 세포 내용물의 품질과 양을 비교하기 위한 단일 센터, 전향적 연구
이 전향적 관찰 연구의 목표는 두 가지 세포 자극 단백질인 filgrastim과 pegfilgrastim 중 하나를 투여한 후 수확한 혈소판 풍부 혈장의 세포 내용물의 질과 양을 비교하는 것입니다. 대답하려는 주요 질문은 다음과 같습니다.
• 4일 필그라스팀 치료와 1일 페그필그라스팀 치료를 비교할 때 참가자의 세포 내용물이 비슷합니까?
참가자는 다음과 같은 중재를 받게 됩니다.
- 동의 후 첫 방문과 치료 후 재방문 시 130mL 채혈(필그라스팀 4일, 페그필그라스팀 7일)
- 전체 참가자의 절반이 필그라스팀을 먼저 받고, 필그라스팀 치료가 끝난 후 8주 후에 페그필그라스팀을 받습니다. 나머지 절반은 역순으로 치료를 받게 됩니다.
연구자들은 각 치료 투여 후 세포 내용물의 질과 양을 비교할 뿐만 아니라 어떤 순서로 치료를 받았는지에 따라 데이터의 차이를 비교할 것입니다.
연구 개요
상세 설명
제안된 연구는 10명의 건강한 지원자가 참여하는 전향적, 무작위 통제, 단일 센터 실험실 연구입니다. 잠재적 참가자가 스크리닝을 통과하고, 연구 절차에 동의하고, 의료 면담 및 실험실 혈액 검사를 완료하면 피험자는 두 번의 연속 동원 이벤트를 겪게 됩니다. 동원 이벤트의 일정은 동원 이벤트의 순서 효과에 대응하기 위해 10명의 참가자에 따라 다양합니다. 5명의 건강한 기증자에게 4일 동안 하루에 10mcg/kg의 표준 필그라스팀 동원 요법을 투여합니다. 이후 참가자 5명에 대해 8주 간격으로 6mg 주사로 구성된 표준 페그필그라스팀 동원 요법이 뒤따를 것입니다. 다른 5명의 건강한 기증자는 약제의 역순으로 먼저 페그필그라스팀 다음에 필그라스팀을 투여합니다.
연구 첫날에 실험실 테스트를 위한 표준 PRP를 생성하는 데 사용되는 130mL의 첫 번째 혈액 채취가 수행되고 피험자는 필그라스팀 또는 페그필그라스팀 투여 시리즈를 시작합니다. 지정된 시간(필그라스팀의 경우 4일, 페그필그라스팀의 경우 7일) 후 두 번째 혈액 130mL를 채취하여 Arthrex Angel 시스템으로 처리하여 실험실 테스트용 M-PRP를 생성합니다. 표준 PRP 및 M-PRP 세포 함량은 세포 계수, 세포 배양 및 단백질 분석을 통해 시험관 내에서 연구되고 정량화됩니다. 두 번째 채혈 후 8주 후 피험자는 세 번째 130mL 채혈을 위해 돌아와 두 번째 동원제(페그필그라스팀 또는 필그라스팀)를 투여합니다. 지정된 시간(필그라스팀의 경우 4일, 페그필그라스팀의 경우 7일) 후 환자는 130mL의 네 번째 채혈을 위해 다시 방문합니다. 샘플은 Arthrex Angel 시스템으로 처리되어 실험실 테스트용 M-PRP를 생성합니다. M-PRP 제품의 세포 함량은 세포 계수, 세포 배양 및 단백질 분석을 통해 시험관 내에서 연구되고 정량화됩니다. 그 후 M-PRP 제품의 세포 함량을 필그라스팀과 페그필그라스팀 동원제 간에 비교합니다.

- 19~39세의 건강한 50~100kg
- 피험자는 필그라스팀 치료 및 추가 채혈을 위해 5일 연속 방문하고, 8주 후 페그필그라스팀 치료를 위해 2회 추가 방문하고 7일 후 채혈하는 데 동의합니다. 이 관리 순서는 참가자의 절반에게 제공되며 나머지 절반은 역순으로 동일한 치료를 받게 됩니다.
