Detaljeret beskrivelse
Denne undersøgelse er et randomiseret, placebo-kontrolleret forsøg, hvor forsøgspersoner, der er tildelt den modificerede fastekohorte, vil gennemgå på hinanden følgende ugentlige cyklusser med modificeret faste i to på hinanden følgende dage.
Det primære formål er at vurdere virkningerne af gentagne perioder med modificeret faste på kropsvægten.
De sekundære mål omfatter vurdering af virkningerne af modificeret faste på procent kropsfedt, taljeomkreds og fasteniveauer af hs-CRP (hs-C-reaktivt protein), IL-6 (Interleukin-6), IL-8 (interleukin-8) ), IL-12 (interleukin-12), TNF-a (tumornekrosefaktor-alfa), IGF-1 (insulinlignende vækstfaktor 1), IGFBP-1 (insulinlignende vækstfaktorbindende protein 1), i alt Kolesterol, LDL-kolesterol, HDL-kolesterol, Glukose, Triglycerid, Insulin, Vitamin-D, 25-Hydroxy,
Deltagerne vil gennemgå vurderinger af blodprøver, vitale tegn, BMI, kropsvægt, taljeomkreds og procent kropsfedt.

Inklusionskriterier:
- Ambulant, mand eller kvinde, 21-65 år
- Et kropsmasseindeks (BMI) på 23-34,9
- Generelt sund og uden væsentlige problemer med fordøjelsen af maden
- Har generelt været vægtstabil i de sidste seks måneder
- Er villig til at afbryde brugen af ethvert (ikke-studie) multivitamin- eller D-vitamintilskud under undersøgelsen
- Villig og i stand til at give skriftligt informeret samtykke
- Forstår klart procedurerne og studiekravene
- Villige og i stand til at overholde alle undersøgelsesprocedurer, herunder at følge anbefalingerne for at opretholde deres sædvanlige kost og regelmæssige aktivitet, i henhold til protokol
- Kunne kommunikere, herunder læse, på engelsk
Primære ekskluderingskriterier:
- At have røget cigaret, elektronisk cigaret, cigar, pibe eller brugt et rekreativt stof inden for de seneste 30 dage
- Anamnese med allergi eller følsomhed over for enhver komponent i undersøgelsesprodukterne, herunder mælk, soja og mandler
- Donation af blod med 30 dage før screening/baseline
- Manglende evne til at give en venøs blodprøve
- Deltagelse i en anden undersøgelse inden for 30 dage før baseline/screening
- At være gravid eller planlægge at blive gravid under studiedeltagelse; eller amning
- At være blevet diagnosticeret, modtaget medicinsk behandling eller taget medicin dagligt for følgende medicinske tilstand(er):
- Diabetes mellitus
- Spiseforstyrrelse
- Akut eller kronisk inflammatorisk sygdom eller autoimmun sygdom
- Kardiovaskulær sygdom
- Mave-tarmsygdom, herunder galdeblæreproblemer, galdesten eller galdevejsobstruktion
- Skjoldbruskkirtelsygdom (medmindre på en stabil dosis af medicin i 3 måneder før screening og usandsynligt at ændre medicin eller dosis under undersøgelsen)
- Hypertension (medmindre på en stabil dosis af medicin i 3 måneder før screening og usandsynligt at ændre medicin eller dosis under undersøgelsen)
- Psykiatrisk lidelse
- Neurologisk tilstand/sygdom
- Kræft (medmindre anden hudkræft end melanom, som er blevet behandlet > 3 år før baseline/screening)
- Aktiv eller kronisk lever-, bugspytkirtel- og nyresygdom
- Blodkoagulationsforstyrrelse
- Anden tilstand eller medicinbrug, der ville udelukke deltagelse i undersøgelsen efter investigatorens/sub-investigatorens bedømmelse (Sub-I)
- I øjeblikket tager eller har taget inden for de 30 dage før screening/baseline enhver hormonsubstitutionsterapi (inklusive DHEA (dehydroepiandrosteron), østrogen, progesteron eller testosteron; undtagen dem, der anvendes som præventionsmetode, og som er blevet taget i > 3 måneder uden forventet ændring i undersøgelsens varighed)

- At have haft et kirurgisk indgreb eller have et internt medicinsk udstyr, som efter PI/Sub-I's vurdering ville udelukke deltagelse i undersøgelsen
- Har unormale laboratorietestværdier for screening, herunder bilirubin > 2,5 x ULN (øvre grænse for normal), AST/SGOT (aspartataminotransferase/serumglutamin oxaloeddikesyretransaminase) og ALT/SGPT (alaninaminotransferase/serumglutamin pyrodruevintransaminase) > ULN2,5. kreatinin > 1,5 mg/dL, blodsukker < 85 mg/dL eller > 125 mg/dL og TSH (thyroidstimulerende hormon) > 4,12 µIU/mL eller andre laboratorietestresultater, der ville udelukke undersøgelsesdeltagelse i bedømmelsen af PI/Sub-I
- At have blodtryksmålinger ved baseline/screening > 140 systolisk eller > 90 diastolisk på to på hinanden følgende aflæsninger, medmindre det er tilladt at fortsætte til næste besøg efter PI/Sub-I's vurdering
- Indtager i øjeblikket mere end 7 standard alkoholholdige drikkevarer om ugen for kvinder og 14 drinks om ugen for mænd (en standard alkoholholdig drik er defineret som en flaske/dåse øl, et glas vin eller en ounce hård spiritus)
- Kan eller vil ikke undgå at indtage grapefrugtjuice eller frisk grapefrugt, Sevilla-appelsiner og tangelos
