De HE10A/13 studie is een observationele studie van de Hellenic Cooperative Oncology Group (HeCOG). Het belangrijkste doel is het verzamelen van biologisch materiaal van patiënten op zoek naar prognostische / voorspellende biomarkers. Elke patiënt ondertekende een geïnformeerde toestemming voor het verstrekken van biologisch materiaal voor toekomstige onderzoeksdoeleinden. De studie werd uitgevoerd in overeenstemming met de Verklaring van Helsinki. Het klinische en translationele protocol is goedgekeurd door het Wetenschappelijk Comité van de Hellenic Cooperative Oncology Group en wordt beoordeeld door de Institutional Review Board van het Metropolitan Hospital. Patiënten uitsluitend met luminaal A of luminaal B (oestrogeenreceptoren (ER)-positief met of zonder progesteronreceptoren (PgR) positief, HER-negatief) borstkanker die adjuvante chemotherapie kregen met positieve lymfeklieren of lymfeklier-negatief met tumorstadium of pathologische kenmerken volgens de richtlijnen van Saint Gallen die een hoog risico suggereren of wanneer genomische testen worden bepaald in het voordeel van adjuvante chemotherapie, in aanmerking komen voor registratie in het onderzoek.
Vóór registratie voor het onderzoek ondertekende elke patiënt een geïnformeerde toestemming voor het verstrekken van biologisch materiaal voor onderzoeksdoeleinden.
Potentiële biomarkers zullen worden onderzocht met behulp van in situ of moleculaire methoden in in formaline gefixeerde, in paraffine ingebedde (FFPE) tumorcoupes. Voor tumorsubtypering zal immunohistochemie voor ER, PgR, HER2, Ki67 en Fluorescentie in Situ Hybridisatie (FISH) worden uitgevoerd waar nodig voor de beoordeling van de HER2-status; voor epidermale groeifactorreceptor (EGFR) en CK5 voor de classificatie van basaalachtige tumoren; en de expressie van CD8, CD3 en FOXP3 op (tumorinfiltrerende lymfocyten (TIL's) zal ook worden geëvalueerd. Bovendien zal het mutatieprofiel worden geëvalueerd, inclusief bruikbare genomische veranderingen, en het immuunresponsgerelateerde profiel van Griekse vrouwen met borstkanker, via de toepassing van DNA / RNA Next Generation Sequencing (NGS) -technologieën, ook in relatie tot de patiëntuitkomst.
