En fase 3 randomisert, dobbeltblind studie av nivolumab monoterapi eller nivolumab kombinert med ipilimumab vs placebo hos deltakere med lokalisert nyrecellekarsinom som gjennomgikk radikal eller delvis nefrektomi og som har høy risiko for tilbakefall

Hensikten med denne studien er å fastslå om nivolmab alene eller kombinasjonen av nivolumab og ipilimumab versus placebo, er trygt og effektivt for å forsinke eller forhindre tilbakefall av kreft hos deltakere som har opplevd delvis eller fullstendig fjerning av en nyre.

Inklusjonskriterier:

  • Nyretumor er fullstendig resekert med negative kirurgiske marginer. Randomiseringen må skje mer enn 4 uker og mindre enn (eller lik) 12 uker fra datoen for nefrektomi
  • Patologisk svulst-, node- og metastase-stadie (TNM) som møter en av følgende: pT2a, G3 eller G4, N0 M0; pT2b, G hvilken som helst, NO M0; pT3, (a, b, c), G hvilken som helst, NO M0; pT4, G hvilken som helst, NO M0; pT hvilken som helst, G hvilken som helst, N1 M0
  • Svulst etter nefrektomi viser nyrecellekreft (RCC) med en overveiende klar cellehistologi, inkludert deltakere med sarkomatoide egenskaper
  • Deltakere må ikke ha kliniske eller radiologiske bevis på makroskopisk gjenværende sykdom eller fjernmetastaser etter nefrektomi
  • Eastern Cooperative Oncology Group (ECOG) ytelsesstatus (PS) 0-1
  • Kvinner må godta å følge prevensjonsmetoder, hvis det er aktuelt

Ekskluderingskriterier:

  • Deltakere med en aktiv kjent eller mistenkt autoimmun sykdom
  • Kjent historie med positiv test for humant immunsviktvirus (HIV) eller kjent ervervet immunsviktsyndrom (AIDS)
  • Tidligere behandling med et anti-PD-1-, anti-PD-L1-, anti-PD-L2- eller anti-CTLA-4-antistoff, eller et hvilket som helst annet antistoff eller medikament som er spesifikt målrettet mot T-celle-kostimulering eller sjekkpunktveier
  • Enhver alvorlig eller alvorlig, akutt eller kronisk medisinsk eller psykiatrisk tilstand, eller laboratorieavvik som kan øke risikoen forbundet med studiedeltakelse
  • Anamnese med allergi eller overfølsomhet for å studere legemiddelkomponenter
  • Deltakere med en tilstand som krever systemisk behandling med kortikosteroider
  • Deltakere som har fått en levende/svekket vaksine innen 30 dager etter første behandling
