Detaillierte Beschreibung
In dieser randomisierten Phase-II-Studie soll die Wirksamkeit und Toxizität von Nimotuzumab in Kombination mit einer Radiochemotherapie bei inoperablem, lokal fortgeschrittenem Plattenepithelkarzinom der Lunge bestimmt werden.
Für alle Patienten war eine radikale Bestrahlung des Brustkorbs und gleichzeitig eine wöchentliche Chemotherapie mit Docetaxel und Cisplatin über jeweils einen Tag vorgesehen.
Die Nimotuzumab-Gruppe wurde wöchentlich mit Nimotuzumab (200 mg, i.v.) in Kombination mit einer Radiochemotherapie behandelt, während die Kontrollgruppe mit einer Radiochemotherapie behandelt wurde.

Beschreibung
Einschlusskriterien:
- histologisch bestätigter Plattenepithelkarzinom der Lunge
- Patienten haben messbare oder auswertbare Läsionen auf der Grundlage der RECIST-Kriterien (Response Evaluation Criteria in Solid Tumors).
- inoperabler Lungenkrebs der Phasen IIIA (N2) und IIIB, bestätigt durch CT oder MRT
- ECOG-Leistungsstatus 0-1
- Zuvor mit Chemotherapie behandelt oder therapienaiv
- Keine vorherige Strahlentherapie, Immuntherapie oder Biotherapie des Brustkorbs.
- Hämoglobin ≥ 10 mg/dL, Blutplättchen ≥ 100.000/μL, absolute Neutrophilenzahl ≥ 1.500/μL
- Serumkreatinin ≤ 1,25-facher oberer Normalwert (UNL) oder Kreatinin-Clearance ≥ 60 ml/min
- Bilirubin ≤ 1,5-fach UNL, AST (SGOT) ≤ 2,5-fach UNL, ALT (SGPT) ≤ 2,5-fach UNL, alkalische Phosphatase ≤ 5-fach UNL
- FEV1 >0,8 L
- CB6 innerhalb normaler Grenzen
- Patienten und ihre Familien unterzeichneten die Einverständniserklärung
Ausschlusskriterien:
- adenosquamöses Karzinom
- frühere oder kürzlich aufgetretene andere bösartige Erkrankungen, mit Ausnahme von nicht-melanozytärem Hautkrebs oder Gebärmutterhalskrebs in situ
- Kontraindikation für eine Chemotherapie
- Frauen in der Schwangerschaft, Stillzeit oder ohne Schwangerschaftstest 14 Tage vor der ersten Dosis
- Frauen, bei denen die Wahrscheinlichkeit einer Schwangerschaft ohne Verhütung besteht
- Blutungsneigung
- in anderen klinischen Studien innerhalb von 30 Tagen
- drogen- oder alkoholabhängig, AIDS-Patienten
- unkontrollierbare Anfälle oder psychotische Patienten ohne Selbstkontrollfähigkeit
- schwere Allergie oder Eigenart
- Nach Einschätzung der Forscher nicht für diese Studie geeignet
