O objetivo do estudo é avaliar a eficácia do tratamento contra o vírus da hepatite D (HDV) do regime JNJ-73763989 + análogo de nucleos(t)ide (NA) em comparação com NA sozinho.

JNJ-73763989 é um terapêutico antiviral direcionado ao fígado para injeção subcutânea projetado para tratar a infecção crônica pelo vírus da hepatite B (HBV) por meio do mecanismo de interferência do ácido ribonucleico.
Este estudo de fase 2 é projetado para avaliar a segurança e eficácia de JNJ-73763989 em pacientes infectados por HBV que são co-infectados com HDV.
O estudo consiste em 2 partes: a Parte 1 avaliará a segurança, tolerabilidade e atividade antiviral de JNJ-73763989 + NA, enquanto a Parte 2 avaliará a segurança e eficácia do regime JNJ-73763989 + NA no tratamento da co-infecção HBV/HDV .
Cada parte inclui 3 fases: Fase de triagem (de 4 semanas até no máximo 8 semanas), fase de intervenção (144 semanas para o braço A e 148 semanas para o braço B) e fase de acompanhamento (48 semanas).
A duração da participação individual no estudo será entre 196 e 204 semanas.
Segurança e tolerabilidade (incluindo eventos adversos [EAs] e EAs graves, avaliações laboratoriais, eletrocardiograma [ECG], sinais vitais, exame físico), eficácia (incluindo ácido ribonucleico [RNA] HDV, ácido desoxirribonucleico [DNA] HBV e antígenos) e a farmacocinética será avaliada ao longo do estudo.

Critério de inclusão:
- Medicamente estável com base no exame físico, histórico médico, sinais vitais, eletrocardiograma (ECG) na triagem
- Coinfecção crônica pelo vírus da hepatite B (HBV) e vírus da hepatite D (HDV) com documentação de pelo menos 6 meses antes da triagem
- Para a Parte 1: ARN da hepatite D (ARN do HDV) superior ou igual a (>=) 1000 unidades internacionais por mililitro (UI/mL) no rastreio. Para a Parte 2: deve ter valores de HDV RNA >= 500 UI/mL e deve ter valores de antígeno de superfície da hepatite B (HBsAg) menores ou iguais a (
- Alanina aminotransferase (ALT) maior que o limite superior normal (LSN), mas menor que 10 vezes (LSN)
- Índice de massa corporal (IMC) entre 18,0 e 35,0 quilogramas por metro quadrado (kg/m^2), extremos incluídos
- Medidas contraceptivas altamente eficazes em vigor para participantes do sexo feminino com potencial para engravidar ou participantes do sexo masculino com parceiras com potencial para engravidar

- Anormalidades laboratoriais significativas, conforme definido no protocolo na triagem
- Participantes com histórico de malignidade dentro de 5 anos antes da triagem
- Ritmo sinusal anormal ou parâmetros de ECG na triagem, conforme definido no protocolo
- História ou arritmia cardíaca atual ou história ou evidência clínica de doença cardíaca significativa ou instável

Off-treatment HDV relapse is defined as: 1.
In participants with HDV RNA <LLOQ (i.e., 630 IU/ml) at EOT: Confirmed increase in HDV RNA of > 1 log10 IU/mL above the LLOQ JNJ-3989 off-treatment.
2. In participants with HDV RNA >LLOQ at EOT: Confirmed increase in HDV RNA of > 1 log10 IU/mL from EOT HDV RNA value JNJ-3989 off-treatment.

Percentage of participants with HBV flare (Virologic, biochemical, and clinical) post end of treatment was reported.
Off-treatment is defined as the time period as the periods when the participants do not receive any of the study interventions (JNJ-3989/placebo and NA).
Virologic flare is for participants who are off-treatment and had HBV DNA < LLOQ at the last observed point on all study treatments, and categorized based on the confirmed (i.e., two consecutive values) peak HBV DNA above any of the three thresholds: 20000 IU/mL, 2000 IU/mL and 200 IU/mL.
Biochemical HBV flare is defined as a confirmed ALT and/or AST>=3*ULN and >=3* nadir (i.e.
lowest value observed up to the time point of meeting the biochemical flare criteria).
An HBV clinical flare occurs either when an HBV virologic flare and off-treatment HBV biochemical flare overlap in time or when a HBV biochemical flare starts within 4 weeks following the end of an HBV virologic flare.
